One question always brought up by the latest examination is how did he paint the image since no brushstrokes are visible?  Since DaVinci was also a scientist and inventor he may have used one of the first versions of airbrushing. That would not leave any evidence of stroking and could be applied in many many layers, painstaking of course to get the detail he did, but possible. Naturally he wouldn't have the powered or computer aided versions we have today but instead maybe used his own breath, or perhaps a foot operated pedal and blowers.
